What is Sonidegib?

Sonidegib (Odomzo) is an oral targeted therapy primarily used for locally advanced basal cell carcinoma in adults. It is generally considered when the cancer has recurred after surgery or radiation, or when surgery or radiation are not good treatment options.

How does Sonidegib work?

Sonidegib is a hedgehog pathway inhibitor. It works by blocking a signaling pathway that helps drive the growth of certain basal cell carcinomas.

What are possible side effects of Sonidegib?

Common side effects include muscle spasms, hair loss, changes in taste, nausea, fatigue, diarrhea, weight loss, and abdominal pain. Sonidegib can also cause muscle-related problems and increases in creatine kinase, so providers may monitor for muscle symptoms and lab abnormalities during treatment.

How is Sonidegib used?

Sonidegib is taken by mouth once daily on an empty stomach. Treatment is generally continued until the cancer remits, progresses or side effects make treatment inappropriate. Because this medication is used for advanced skin cancer, it may be managed in coordination with specialists, which may include dermatology, surgical oncology, radiation oncology, or medical oncology depending on the case.

Sonidegib can cause severe birth defects and should not be used during pregnancy. Pregnancy precautions apply to both women who can become pregnant and men with partners who may become pregnant, and contraception precautions continue for an extended period after treatment stops. The prescribing information also advises against donating blood or semen for a prolonged period after treatment.
